Yes, they mean single-core. If it required a dual-core, it would have said so specifically.
2 2.5 ghz should be able to handle it easily enough, particularly since you've got a 9000-series Nvidia backing it up. I guess you're using Windows XP? Stick with that. Shut down as much other stuff as you can though, to give as many of your pc's resources to the game as possible.
You could look into overclocking your cpu's too, but there will be no need here.
Modern intel and amd processors run more efficiently at around 2.0 ghz than above 3ghz. It's much better to have 2 running at 2.5 than 1 running at 2.8.
